Effect of adenosine triphosphate on methanol induced oxidative liver injury in rats

Abstract (EN)
Introduction and Aim: Methanol (methyl alcohol) is a colorless, volatile and toxic type of alcohol. In our country, it has been found that methanol poisoning has been observed at a certain level over the years. Methanol has serious clinical consequences from blindness to death. The primary toxic factor in methanol poisoning is metabolic acidosis. In many studies, methanol has been proven to cause hepatotoxicity due to oxidative damage. Although adenosine triphosphate (ATP) has been shown to be effective in reducing oxidative damage in studies on ischemic tissue, there is no study on the effect of methanol-induced hepatotoxicity on oxidative damage. In this study, to investigate the protective effect of ATP against liver damage caused by methanol in rats. Material and Method: In our study, a total of 18 Wistar albino male rats in the range of 235-245 g were randomly divided into 3 groups. Rats were divided into healthy group (SG), methotrexate + methanol Group (MTM) and methotrexate + methanol + ATP (MMA) groups. No action was taken on the healthy group. After the methotrexate was given orally to the MTM and MMA group for 7 days with a gavage of 0.3 mg / kg, 20% methanol was given to the MTM and MMA groups at a dose of 3 g / kg and in the same route. 4 hours after methanol administration, 4 mg / kg dose of ATP was injected in the intraperitoneal route. Serum ALT and AST measurement in blood samples taken from animals 8 hours after ATP injection, MDA and GSH analyzes and histopathological examinations were performed on liver tissue samples. Results: While ALT, AST values and AST / ALT ratio were observed at the lowest SG, it was found to be significantly higher in the MTM group compared to the other two groups, and it was found to be lower than the MTM group in the MMA group. MDA was measured at the lowest SG and the highest MTM group. In the MMA group, it was found to be lower than the MTM group. GSH was found in the highest healthy group, while it was higher in the MTM group than the MMA group. When the tissues are evaluated histopathologically; While hepatocyte degeneration, kupffer cell increase, PMNL infiltration and congestion in vessels were observed in the MTM group, degenerative effects were significantly reduced in the MMA group. Conclusion: In the light of these findings, we believe that ATP has a protective effect against liver damage caused by methanol, but more studies should be done in terms of applicability in these indications in the clinic.
