Investigation of the effect of meningich (pistacia terebinthus l.) extract on ischemia/reperfusion injury induced by experimental torsion/detorsion in testis

Aim: The aim of this thesis was to investigate the protective and therapeutic effects of meningic (Pistacia terebinthus L.) extract on ischemia/reperfusion injury induced by experimental torsion/detorsion in the testis. Materials and Methods: 48 male Sprague Dawley rats were divided into 6 groups. For the torsion-detorsion procedure, the right and left testes of the animals were rotated 720 degrees and torsion was performed for 3 hours, followed by detorsion for 3 hours. The scrotums of the animals in the sham group were opened with a surgical protocol and closed without any further intervention. The animals in the meningic group were given meningic extract for 15 days different from the sham group. The animals in the torsion-detorsion group were subjected to torsion for 3 hours and detorsion for 3 hours, and the animals were sacrificed. The animals in the meningic+torsion-detorsion group received 2 ml/kg of meningic extract for 15 days before torsion-detorsion, the animals in the torsion-detorsion+meningic group received 2 ml/kg of meningic extract for 15 days after torsion-detorsion, and the animals in the meningic+torsion-detorsion+meningic group received 2 ml/kg of meningic extract for 15 days before and after torsion-detorsion. At the end of the experiment, testes of the animals were removed and histopathologic and immunohistochemical evaluations were performed. Results: In the testicular tissues of animals in sham and meningic groups, regular seminiferous tubules and 5-6 rows of germinal epithelium were observed on the basal lamina. Capillaries and leydig cells in the interstitial area were normal. In the torsion-detorsion group, destruction of seminiferous tubules, deterioration in spermatogenic cells, hemorrhage, edema and congestion in the interstitial area were observed. Partial recovery in seminiferous tubules, dilatation and congestion were observed in the seminiferous tubules of animals in Menengiç+Torsion-detorsion group. In torsion-detorsion+menengic and menengic+torsion-detorsion+menengic groups, basal laminae showed similar characteristics to the sham group and congestion was observed in the vessels. Conclusion: Menengiç extract was found to contribute to the regeneration of testicular tissue against ischemia-reperfusion injury caused by torsion-detorsion in the testis. It was observed that it supported the survival of cells by suppressing Apaf-1 expression in the treatment groups. It was observed that it contributed to the continuation of spermatogenesis process by increasing Sox-9 expression in the treatment groups.
