Abstract (EN)
Pseudomonas aeruginosa, a common cause of hospital-acquired infections, has become challenging for clinicians worldwide because of its decreasing rates of susceptibility to broad-spectrum antibiotics to determine P. aeruginosa resistance to antibiotics and the genes that cause this resistance. The descriptive study conducted on one hundred samples were gathered from the operating rooms and burn ward at Al-Hussein Teaching Hospital in Thi-Qar. The isolates were identified by culturing on MacConkey agar, CHROM agar, performing biochemical tests were done which include catalase test and oxidase test. The sensitivity of P. aeruginosa isolated in the present study to 6 antibiotics, using the diffusion method on the Muller Hinton agar media. The results showed Out of 20 P. aeruginosa bacteria, 16 of the bacteria resistance to piperacillin, 18 resistance to ceftazidime, 16 resistance to tobramycin & ciprofloxacin, 19 resistance to amikacin, & 20 resistance to ceftriaxone. All of 20 P. aeruginosa isolates serotyped by slide agglutination test with commercial antisera. 8 serotypes were recognized in specimens as followed O1 was 2 (10%), O2 was 2(10%), O3 was 1(5%), O4 was 2(10%), O6 was 3(15%), O8 was 2(10%), O9 was 2(10%), O11 was 6 (30%), and O11 is often the most prevalent serotypes reported in the presented study was 6 (30%). The sequencing result of algD gene shows for P. aeruginosa having one variation (Transition) A>G code ACT/GCT. The sequencing result of lasB gene shows for P. aeruginosa having one variation T>C code TGA/CGA. The sequencing result of lasB gene shows for P. aeruginosa having one variation T > C code TGG/CGG. The phylogenetic tree was analyzed the algD gene (313bp) and the lasB gene (284bp), it was found that this Phylogenetic tree consisted of only one type of P. aeruginosa bacteria.
