Effect of benfotiamine on ovarian and uterine tissues of rats administered trichloroethylene

Trichlorethylene (TCE) is a chlorinated hydrocarbone. Essentially it is used for reduction of metals. TCE exposure usually becomes the water is contaminated with TCE or evaporates methal reduction process. Because of TCE found in adhesives, for flooring cleaning solution, stain remover is a hazardous substance that can be found in the house. Many studies done by TCE showed its toxic effects on liver, kidney and ovarian tissues. Benfothiamin is vitamin B1's fat soluble form. Benfothiamin showed repressive effect on reactive oxygen metabolites. The aim of this study was to investigate the effects of benfothiamin on ovarian and uterus tissue of rats exposed to TCE. In this study, 24 female 8-10 weeks old Wistar-Albino rats were used. The rats were randomly divided into 4 groups. Nothing was given during the 3 weeks test period to control group. It was given 1000mg/kg/day TCE to TCE group, orally. It was given 1000mg/kg/day TCE and 70 mg/kg/day benfothiamin to TCE+Benfothiamin group, orally. It was given 70mg/kg/day benfothiamin to benfothiamin group, orally. After the experimental period the all groups of rats were decapitated under anesthesia. After decapitation removed ovarian and uterus tissues were embedded into paraffin blocks by routine procedures for histologic evaluation. The tissue sections are received from paraffin blocks is stained with Masson Trichrome and examined the histopathologic changes in groups. Also TUNEL technic was applied to this section and established apoptotic cells. However over and uterine tissues is hold in -80 °C for measure MDA (malondialdehid) levels. Ovarian tissues in TCE groups showed significant increase MDA levels, apoptotic cells and histopathologic changes. Compared with TCE group in TCE+Benfothiamin group the changes showed decrease. In the uterine tissue, while MDA levels showed decrease in TCE group, MDA levels showed significant increase in TCE+ Benfothiamin group. There is no important different between groups in evaluation of apoptotic cells. Histopathologically, while inflammatory cells increased in TCE group's uterus tissue, TCE+Benfothiamin group was no changes. Consequently, TCE enhances MDA, apoptotic cells and histopathologic changes in over tissue and treatment of Benfothiamin reduces these changes. After all TCE has made limited changes in uterus, more advanced and extensive studies are needed to designate other effects. Keywords: Trichlorethylene, Uterus, Over, Benfothiamin
