Abstract (EN)
Xanthine oxidase (XO) is a versatile flavoprotein enzyme found in all mammals' tissues (from bacteria to humans), to generate reactive oxygen species (ROS), the enzyme catalyzes the oxidative hydroxylation of purine substrates at the molybdenum centre (the reductive half-reaction) and then reduces O2 at the flavin centre (the oxidative half-reaction). An imbalance of metabolites in the body causes many illnesses, or at least their symptoms. Excess uric acid, for example, may cause gout. Inhibition of XO has an antihyperuricemic effect by lowering uric acid levels. Allopurinol is a therapeutically effective xanthine oxidase inhibitor used to treat gout. The fact that xanthine oxidase blood levels are elevated in pathological conditions such as hepatitis, inflammation, ischemia-reperfusion, carcinogenesis, and aging is well accepted. Thus, inhibiting this enzymatic pathway may be advantageous. This review will summarize recent advances in understanding the enzyme's structure and action, related disease conditions, and attempts to produce novel xanthine oxidase inhibitors.
