Abstract (EN)

The aim of this study was to develop donepezil loaded nanoparticle formulations for oral administration. Therefore, donepezil loaded nanostructured lipid carriers comprising coconut oil which has anti-Alzheimer effect prepared by microemulsion method and solid lipid nanoparticles prepared by high speed homogenization method were developed using different solid lipids, oils, surfactants and co-surfactants. Liposome formulations were prepared by using thin-film hydration method. The study continued with optimized nanostructured lipid carriers based on characterization results. Donepezil diffusion from the donepezil loaded ideal formulation was determined by in vitro release study using dialysis membrane. Drug release was determined in stomach and intestine medium. Moreover cell culture and cytotoxicity study were carried out. Physicochemical properties of the ideal formulation were determined by several analyses such as pH, viscosity, particle size, particle size distribution, zeta potantial and stabilities. In the light of these informations, donepezil loaded nanostructured lipid carrier formulation has been considered and suggested to decrease potential side effects as a result of controlled release feature and to be more effective for Alzheimer's disease treatment. Ke words: Al heimer s Disease; nanostructured lipid carriers; liposome; donepezil
