Abstract (EN)
Behçet's Disease (BD) was first discovered in 1937 by Turkish dermatologist Dr. BD is a universal disease, and studies show that Turkey has a high prevalence compared to other cases of BD. Behçet's Disease has become a condition of increasing importance, as it generally affects young adults, is difficult to treat, and causes serious morbidity with organ involvement. Increased reactive oxygen species (ROS) caused by inflammation disrupt the oxidant/antioxidant distribution of the organism and cause damage to biomolecules such as DNA. One of the most common DNA damages caused by ROS in DNA is 8-Oxo-2'- deoxyguanosine (8-oxoG). In Behçet's diseases, when there is no possible disease with repeated treatment, ROS-induced oxidative stress is likely to be high and cause DNA damage. Numerous reports have shown that 8-OHdG detected in biological materials is a good biomarker used to determine existing DNA damage in diseases such as cancer, Parkinson's disease, systemic lupus erythematosus, rheumatoid arthritis. Repair of 8-OHdG damage in DNA occurs with the OGG1 enzyme, which is involved in the base excision repair (BER) pathway. The aim of our study is xvi to reveal possible DNA damage in BD and to reveal the reduction of DNA damage in the pathogenesis of BD by identifying the OGG1 gene that plays a role in the repair of this damage. The software we developed detected OGG1 gene expressions with real-time polymerase chain properties (qPCR; qPCR). Using the ELISA method to use the amount of 8-OHdG, which is an indicator of oxidative DNA infection. The studies were conducted in a total of 70 BDs and 30 healthy controls of similar age and gender. In terms of changes in OGG1 mRNA, the opposite relationships between the control group and the entire BD group, no significant difference was observed between the two groups (p = 0.20). Similarly, no significant difference was detected between the control group and the problems of the D-BH (Deep Vein Thrombosis-BD) group (p = 0.59) and M-BH (Mucocutaneous-BD) group (p = 0.18). On the other hand, a difference was detected between the D-BH (0.0040±0006) and M-BH (0.0024±0.0002) group sales against the two groups, where the OGG1 mRNA group was kept at a strategic point (p=0.024). ; OGG1 expression level was found to be lower in the M-BH group. No significant difference was observed in 8-OHdG levels between the disease control group and the entire GH group (p = 0.27). Similarly, no difference was detected in maintaining 8- OHdG levels between the D-BH group (p = 0.42) and the M-BH group (p = 0.28) compared to the control group. There was no significant difference in 8-OHdG point between the D-BH group and the M-BH group (p = 0.85). This is the first study in the literature investigating the relationship between DNA repair rates and Behçet's disease, and may provide many guidance in this field.
