Abstract (EN)
Defining the best postcondition time in skeletal muscles to minimize the ischemia reperfusion injury Purpose:After reperfusion of tissues which were exposed to ischemia develop wide cell necrosis due to oxygen radicals. According to the literature poscondition minimizes the effect of ischemia reperfusion injury. In our study we compared not only the postcondition effect but also we compared the best interval times to protect from ischemia reperfusion. Methods: 20 adult Sprague ? Dawley rats were used in this study and randomized into 5 groups. Before the surgical prosedure we anestezied rats with 50mg/kg ketamine and 5mg/kg xylazine. Bilateral inguinal dissection was performed and the femoral arteries were diseccted. A microvascular clemp was used to inhibit the artery flow for four hours. In the control group the vascular clemp was removed directly without any controlled reperfusion whereas in the study groups the clamp was removed and controlled reperfusion was performed for 5,10,15 and 20 sec (According to the study groups) while controlled reperfusion was performed. The rat were sacrified and the gastrokinemus muscle exized, and NO,SOD levels and viability percentage was compared. Findings: NO levels (mean 0,131 µM/wet tissue) were significantly lower in the 15 sec study group compared to the other groups (p<0,05). Also SOD levels (mean 351,11ng/gr wet tissue) were significantly low in the 15 sec study group (p<0,05). Comparing all groups the viabilty of 15 sec group was the highest. Conclusion:According to the results of our study controlled reperfusion of 15 seconds minimizes the injury from ischemia reperfusion more than 5, 10 and 20 sec postconditioning.
