Abstract (EN)

ABSTRACT THE PROTECTIVE ROLE OF RIFAXIMIN IN NON ALCOHOLIC FATTY LIVER DISEASE CREATED WITH FRUCTOSE Non-alcholic faty liver disease isthe most frequent liver disease in the worldwide. In addition it is commonly interested with the methabolic syndrome. There is possibility that the disease may be associated with the increase fructose consumption. In this study, which will be created with fructose in an experimental model of fatty liver, we aimed to investigate the protective effects of rifaksiminThere are42 male Sprague-Dawley rats in the study. They were divided into 6 equal groups according to following feed types: Control group; given the 50% fructose solution; given one time rifaksimin per week with fructose solution; given three times rifaksimin per week with fructose solution; normal diet with given one time rifaksimin per week; normal diet with given three times rifaksimin per week. After eight weeks, the tissue samples, getting from rat liver, were taken for histopathological examination and levels of MDA, TNF-α, Collagen type 1, NF-kappaB, Nrf-2 ve HO-1.Blood samples were taken forbiochemical examination and levels of plasma Glutatyon peoksidaz, TNF-α, 4-hydroxynonenal. It was seen in histopathological examination that the develeopment of ballooning degeneration, inflamation and macrovesicullar steatosis which they are the findings of hepatic steatosis in the group of feeding with 50% fructose solution. On the other hand, he meaningful decreasing in the findings of hepatic steatosis was observed in group of taking % 50 fructose solution with rifaksimin. The level of TNF-α of plasma and tissue, NF-kB, MDA, type 1 collagen level, plazma 4-HNE were observed as low in the group taken rifaksimin placed with patogenesis of hepatosteatosis. However, levels of GSH-Px, Nrf-2 HO-1 observed as high in treatment group. In the group receiving high doses of Rifaksimin did not differ significantly. Rifaksimin; lubrication, ballooning degeneration andinflammation were reduced in fructose diet NAYK model created. It was thought that rifaksimin prevent the steatohepatitis providing oxidatif stress with intestinal decontamination. New studies on this subject are needed.
