The effects of cherry laurel, lipoic acid and routine flavonoid on metformin-treated diabetic rats

Abstract (EN)
The aim of this thesis is to discover the interactions between the combination of medicine and natural products. For this reason, metformin, which is widely used in type 2 diabetes mellitus, was selected, and 3 different natural products were selected to see the drug interaction, these are cherry laurel extraction, rutin flavonoid and alpha lipoic acid. Before starting the experimental study, the animals were divided into 6 groups, and with the exception of the first group (healthy control), type 2 diabetes was established in the other groups with STZ and NA. The drug and natural products were administered orally for a period of 12 weeks, and when the experiment was done, the animals were sacrificed and their blood and tissues were taken. Serum Biochemistry, HbA1c and Fructosamine values were measured by autoanalyzer, antioxidant enzymes and TAS/TOS, Nitric oxide were measured by the specified methods, insulin and aldose reductase were measured by the ELISA method. In addition, the pancreas tissue was also evaluated from a diabetic point of view by histopathological examination. After discovering the results statistically, they were compared with different studies and it was found that blood sugar levels in particular were statistically insignificant compared to other studies and generally remained close between groups (p>0.05). Statistically significant differences were found in weight changes and Calcium, Ceruloplasmin and ALP values on the day of sacrification. The group with the lowest blood sugar levels was the group given a combination of metformin and cherry laurel, and it was found 130.80 + 21.75 mg/dL, while the group with the highest glucose level was the group given rutin flavonoids and metformin, and it was found 165.89 + 48.89 mg/dL. Only ceruloplasmin values were found to be statistically significant among antioxidant-acting enzymes and compounds, and the highest concentration was observed in the diabetes group, and the lowest was observed in the diabetic group using metformin and routine flavonoids. There was no statistical significance in fructosamine and HbA1c values. Nitric Oxide was statistically significant in brain, kidney and plasma tissues. The results regarding Aldose Reductase are not statistically significant. In the measurement of total antioxidant capacity, no significance was found in other tissues except the eye, and in the analysis of total oxidant capacity, no significance was found in any tissue. The oxidative stress index in serum was the lowest in the control and cherry laurel group, while the highest was found in the diabetic group. More detailed research is needed to better express how drugs and natural products change the effects of each other. More support should be given to studies to increase in vivo studies on drug interactions and to carry out research by applying advanced analysis methods. Diabetic pathways should be examined in more detail and developed with histopathological studies and immunostaining.
