Abstract (EN)

ABSTRACT Analysing the Clinical and Prognostic Values of c-Met, EGFR, PTEN, PDGFRalpha, PI3K/AKT and mTOR Markers in Cases with Malignant Pleural and Peritoneal Mesothelioma Objective: The aim of this study was retrospectively investigation of c-Met, EGFR, PTEN, PDGFR-alfa, PI3K/AKT and mTOR molecules and determination the relationship between clinical and survival changes in malignant pleural and peritoneal mesothelioma cases which were treated and followed up in Cukurova University Faculty Of Medicine Balcalı Hospital Oncology Clinic. Materials and Methods: 53 patients who were admitted to Cukurova University Faculty Of Medicine Balcalı Hospital Oncology Clinic between 2004-2015 and diagnosed with malignant mesothelioma were included to our study. The patients were investigated about demographic characteristics, asbestos exposure, smoking status, tumor site, histological type and stage. As pleural and peritoneal patients were randomized into two groups surgical treatment, radiotherapy and chemotherapy protocols were evaluated. Demographic characteristics of the patients , the treatments they were received and expression levels of c-Met, EGFR, PTEN, PDGFR-alpha, PI3K / AKT and mTOR which were determined by immunohistochemically were evaluated for the effect of overall survival and disease-free survival. Findings: 53 patients were included to the study . Thirty-nine (%73.6) were diagnosed as pleural mesothelioma and other 14(%26.4) were diagnosed as peritoneal mesothelioma . The median overall survival of pleural mesothelioma cases were 15 months and peritoneal mesothelioma cases has been found as 12 months ; moreover there were no significant differences between two groups in terms of overall survival (p = 0.559). The survival of (2+ ; 3+) group were superior to the (0 ; 1+) group when disease free and overall survival were demonstrated depending on c-met and mTOR expression in peritoneal cases (p<0.05) The survival of (0 ; 1+) group were superior to the (2+ ; 3+) group when disease free and overall survival were demonstrated depending on PDGFR-alpha expression in pleural cases (p<0.05). Furthermore , the (2+) group was superior to the (0) group depending on EGFR mutation in pleural MM cases.(p<0,05).There were no significant differences between overall and disease free survival groups depending on PTEN and PI3K/AKT expressions both with pleural and peritoneal cases (p>0,05). Results: We thought , analysis of the clinical and prognostic values of c- Met,EGFR,PDGFR-alpha, and mTOR markers in patients with malignant pleural and peritoneal mesothelioma cases may be important on determining the prognosis and new therapy modalities.
