Abstract (EN)

Objective: In this study, we aimed to evaluate the relationship between the development of thrombosis with JAK2V617F mutation.Patients and Methods: We reviewed 106 Philadelphia negative myeloproliferative neoplasms (MPN) diagnosed between 1994-2011 in our clinic to assess the risk of thrombosis associated with JAK2V617F mutation. Median age was 63 years (range 26-90) with 59 (55%) males and 47 (44%) females. Patient characteristics among myeloproliferative neoplasia groups are demonstrated in table 1. Venous thrombosis occurred in 20 patients (18.9 %): Deep Vein Trombosis (DVT) 14, Pulmonary Embolism (PE) 1, DVT+PE 2, Portal Vein Thrombosis 1, Portal+Splenic vein thrombosis 2. Arterial thrombosis occurred in 34 (32,1 %) patients: Myokardial Infarction (MI) 4, Coronary Arterial Disease (CAD) 10, Cerebrovascular Disease (CVD) 5, Peripheral Arterial Occlusion 1, CVD+MI 6, CAD+CVD 7, CAD+PAO+CVD 1. Thrombosis distribution according to jak status in polycythemia vera (PV) patients failed to demonstrate any statistical significance: venous thrombosis, in 4 jak2(+) and 2 jak2(-), arterial thrombosis in 6 jak2(+) and 3 jak2(-), arterial and/or venous thrombosis in 7 jak2(+) and 4 jak2(-) ( OR: 1.44 p=0.694, OR:1,5 p=0.613, OR: 1.283 p=0.736 consecutively). Thrombosis distribution in essential thrombocytosis showed a statistical significance in terms of both venous and arterial thrombosis: venous thrombosis in 12 jak2(+) and 1 jak2(-) , arterial thrombosis in 18 jak2(+) and 5 jak2(-), arterial and/or venous thrombosis in 22 jak2(+) and 5 jak2(-) (OR:12.46 p=0.005, OR:4.140 p=0.013, OR:6.325 p=0.001 consecutively).Results: JAK2V617F mutant status may be useful for predicting thrombosis in myeloproliferative neoplasms.Key words: JAK2 mutant, thrombosis, myeloproliferative neoplasms.
