Effects of different types of calorie restriction application on inflammatory parameters in MMTV-TGF-α mice at different ages

Abstract (EN)

This Article examines the effects of different calorie restriction methods on cytokine levels and the relation between different calorie restriction methods on aging, according to the research on 115 mice. The mice are the MMTV-TGF-α type, female and that spontaneously develop breast tumours during the post-menopausal period. The serum cytokine levels of mice that have been subject to chronic calorie restriction (CCR) and intermittent calorie restriction (ICR), in the ranges of 10, 17 and 50 weeks are assessed. The calorie restriction has been applied with reference to weekly feed consumption of the control group which is named as ad-libitum (AL) group. The calorie restriction rates have been determined as 15 % for the CCR group and 60 % for ICR group, compared to the AL group. The calorie restriction on the CCR group has been periodically progressed, up to the end of the research, as one-week calorie restriction and three-weeks AL feeding. During the process, the mice have been taken blood by the retro-orbital method, and the serum samples have been produced. The levels of IL-1α, IL-6, IL-10, IL-13, TNF-α and MCP-1 have been tested, from the serum samples, by the Multiplex Elisa method. Statistical analyses have been carried out by the Windows version of GraphPad Prism 7.0. As a result of the research, it is established that the CCR feeding model causes the decrease (p>0,05) in the levels of pro-inflammatory cytokines such as IL-1α, IL-6, IL-13 whereas it increases (p>0,05) the level of the anti-inflammatory IL-10, during the range of 10-17 weeks as the first period of ageing. It is also revealed that the ICR feeding model results in similar consequences (p>0,05) during the range of 17-50 weeks as the second period of ageing. Consequently, although it is established that the calorie restriction can change inflammatory responses, more researches are needed due to the statistical non-significance of data obtained and for the purpose of better understanding of which calorie restriction type represses the inflammatory responses.
