The effect of cerium oxide on the liver in sevoflurane applied rats

Abstract (EN)

Aim: In this study, we evaluated the effect of cerium oxide on liver tissue in rats treated with sevoflurane. Materials and Methods: After the ethics committee approval, 24 rats were randomly divided into 4 groups of 6. Group K: Control Group, Group S: Sevoflurane Group, Group CeO2: Cerium Oxide Group, Group S+CeO2: Sevoflurane+Cerium Oxide Group. Sevoflurane was applied to the sevoflurane group with 2% concentration of sevoflurane and 100% oxygen at 4L/min for 2 hours. Cerium oxide was administered through the tail vein at a dose of 0.5 mg/kg 2.5 hours before the procedure. Sevoflurane+ CeO2 group was administered sevoflurane for 2 hours at a concentration of 2% with 4L/min 100% oxygen, 30 minutes after cerium oxide at 0.5 mg/kg dose was administered through the tail vein. Afterwards, ketamine and xylazine were administered intramuscularly to all rats, and blood was taken from their aortas and their liver tissues were removed. Hgb, Hct, ALT, AST, LDH were studied in the blood samples. Liver tissues were examined histopathologically by light microscopy. Results: Hydropic degeneration, lymphocyte and neutrophil infiltration were more common in all groups than in the control group. Hydropic degeneration was found to be significantly lower in the CeO2 group compared to the sevoflurane group. Neutrophil infiltration was significantly lower in the CeO2 group compared to the sevoflurane group. Necrosis and pleomorphism were similar between groups. AST level was found to be significantly higher in sevoflurane group compared to control group. In addition, although the CeO2 and sevoflurane+CeO2 groups were lower than the sevoflurane group, it was not statistically significant. LDH level was found significantly higher in sevoflurane group compared to control group. Also it was found to be significantly lower in the sevoflurane+CeO2 group compared to the sevoflurane group. ALT, Hgb and Hct were similar between groups. Conclusion: Histopathological and biochemical changes were detected in liver in the sevoflurane group. In histopathological liver damage caused by sevoflurane, CeO2 administration may be useful in preserving the hepatic structure. We think that more meaningful and positive results will be obtained in new CeO2 studies that will be done by differentiating the experimental groups in terms of age, gender, and examining biochemical parameters of oxidative damage with repeated exposure to sevoflurane.
