Abstract (EN)

Systemic sclerosis (SSc) is an autoimmune chronic disease that causes fibrozis on the skin and innards. It is thought that oxidative stress has an important role in the pathogenesis. There have been many studies to develop antioxidant therapy methods. Dietary antioxidants, vasodilator therapies, inhibition of xanthine oxidase were tested but they were ineffective or the results were contradictory. In literature there has been no study to evaluate the immunosuppresants' effects on the oxidative stress and disease activity of the SSc. In our study we arranged to measure levels of total oxidant status (TOS), total antioxidant status (TAS) and oxidative stress index (OSI) before and after six months treatment with cyclophosphamide. We aimed to evaluate the role of oxidative stress in the pathogenesis of SSc, its relationship with disease activity and the effects of immunosuppressive treatment on oxidative stres.We herein evaluated 26 early SSc?ed patients of which 22 were women and 4 were males `who have not previously received treatment?. 15 of the patients were diffuse,11 of them were the type of limited. There were 26 female volunteers in the control group.14 patients were treated orally, the other 12 patients were treated with intravenous cyclophosphamide. The vasodilator and steroid therapies were given to the essential patients.TOS and OSI values were very high compared to the control group (p=0.001, p=0.005) before treatment. TOS, OSI and Valentini activity scores showed significant decreases after treatment (p=0.001, p=0.004, p=0.007).After immunosuppressive therapy, as a result of the significantly decrease on TOS and OSI values, cyclophosphamide might be useful in SSc by reducing the impact of early increased oxidative products on cells and tissues. Valentini disease activity score was decreasing in parallel with TOS and OSI's values so it can be considered to use TOS and OSI?s values as a marker of disease activity.
