In vitro micropropagation of Tunceli garlic (Allium tuncelianum (Kollman), N.Özhatay, D.Matev, Ş.Şiraneci)

Turkey is very rich in plant species due to it location on Euro-Siberian, Mediterraneanand Iran ?Turan phytogeographic regions. It has 12054 plant species, out of which about32.56% species are endemic to Turkey. The plants belonging to Allium species have specialplace among these. Turkey has 159 Allium species out of which 53 are endemic to Turkey.Allium tuncelianum studied in this thesis is placed among vulnerable plant species in the RedBook of Turkish Plants. Previous studies show un-successful results from micro propagation,tissue culture or adventitious bulb regeneration from this plant. It is very important to conservethis important plant species. Complete bulb sterilisation was achieve during 100% commercialbleach for 30 minutes. A comparison of the effects of different concentrations of BAP-NAA,2,4-D on longitudinally sectioned ½ and ¼ bulb explants showed leaf regeneration on adaksialside of the explants. No regeneration was recorded on leaf tip and petiole explant. Rootexplants showed bulb regeneration on root tips on MS medium containing variousconcentrations of BAP-NAA. The best bulb regeneration was achieved on MSmedium containing 5 mg/l BAP-0.5mg/l NAA. This methodology test is the prospects of Tunceligarlic production in future for production and breeding purposes.Keywords: Allium tuncelianum, micropropagation, plant growth regulators
