Abstract (EN)

Benign prostatic hyperplasia (BPH) is the most common benign adenoma in men, which leads to caused occlusion on the exits of the bladder. Also BPH is a complex syndrome that causes significant chronic symptoms associated with chronic inflammation and prostate enlargement. Approximately 40 % of men have the risk of developing this disease throughout their lives. BPH is histological characterized by the proliferation of prostatic epithelial and stromal cells and also it occurs due to complex cellular changes which include alterations in proliferation, differentiation and senescence. It was thought that boron and its derivatives could be used as potential treatment of diseases that were characterized by chronic inflammation and cell proliferation such as BPH because of anti-inflammatory and immune response regulatory properties. However it is shown that boron has an anti-carcinogenic property in addition to anti- proliferative effect from the results of epidemiological, animal and laboratory studies. In this respect one of the most important results of this study that it reveals the need for further examined the relationship between boron exposure and BPH. Boron and its derivatives apparently has diverse effects through influencing a cell signaling system or the formation and/or activity of an entity involved in many biochemical processes and these effects increase the likelihood of becoming a target molecule, which prevent the growth of BPH cells. In this context, the boron derivatives in the our study the effects of BPH-1 cell line apoptosis and DNA damage was aimed at determining the molecular terms. Firstly, it is viewed BPH-1 cell line growth kinetics and cytotoxicity test of BA (Boric acid), BO (Boron oxide) and DPB (Disodium pentaborate decahydrate) was made substance. Because DPB gave more effective results among boron derivatives, following assays were carried out by this chemical. Then, pH2AX, pATM and ATM antibodies were examined in Western blot technique in order to investigate the effects of DPB on DNA damage while DNA damage amounts were calculated with pH2AX and pATM foci numbers by immunofluorescent microscope. Finally, apoptotic effects of boron derivatives on BPH-1 cells were also investigated with p53, p-P53, PARP, caspase-3 and caspase-9 antibodies by Western blotting. With this study, it was firstly determined that DPB treatment with BPH-1 cell line leads to apoptotic cell and DNA damage increase in dose and time dependent manner.
