Researching effects of paclitaxel and morinda citrifolia in C6 glioma cells

Abstract (EN)

In this study, in in vitro C6 glioma cell model, different concentration of Morinda citrifolia (noni) extract and paclitaxel dependent ß-catenin signaling pathway and changes in the cell proliferation are examined immunocytochemically.To ases cell proliferation, S-phase spesific marker bromodeoxyuridin (BrdU) immunocytochemistry was performed. While a decrease in BrdU incorporation was observed after noni extract incubation, there was no S-phase cell after paclitaxel and combined noni extract and paclitaxel applications. In glioma cells, ß-catenin expression was observed across the intracelluler margin of plasma membrane. After incubation with noni extract, no change in ß-catenin expression of existing ß-catenin was noticed. A significant increase was observed in nucleus ß-catenin expression after paclitaxel incubation. Also after incubation with combined noni extract and paclitaxel no change in existing ß-catenin expression was noticed.As a result of our study, it was determined that, the expression of plasma membrane localized cell adhesion molecule ß-catenin was not change by effect of noni extract, and ß-catenin expression was decreased by effect of low dose paclitaxel in nucleus.
