Abstract (EN)

Tuberculosis is an important public health problem especially in developing countries. Multi drug resistant Mycobacterium tuberculosis isolates that are resistant to at least rifampicin (RIF) and isoniazid (INH) are an important problem for control programs. Therefore, to make determination, diagnosis and safe susceptibility tests of MDR-TB isolates; new, fast and inexpensive tests are needed. With the appearence of widespread drug-resistant tuberculosis, World Health Organization (WHO) gives an importance to strengthened the control of tuberculosis and mycobacteriology laboratories, to take care of treating multidrug-resistant tuberculosis patients, to provide adequate antituberculosis medication, prevention of tuberculosis transmission and the development of new antituberculosis drugs and diagnosis tools.Nowadays, convectional, commercial based antibiotic susceptibility tests and rapid molecular diagnosis methods are used for drug susceptibility tests of TB. Nevertheless, long time taking convational methods and the standardization of this methods can cause some problems. Conversely, studies are continuing on new diagnostic methods that are expensive for commercial systems, provide fast results and are easy to standardize. Colorimetric microdilution tests were made using various indicator reagents, antituberculosis drug susceptibility tests in fast and sensitive, resistant tuberculosis can be defined. In the study 42 isolates; DLM sensitivity was found to be 0.5-<0.008 mg/mL, LZD sensitivity was 13 isotylals 0.125 mg/L, 14 isotylals ≤0,008 mg/L and one isotylat resistant. Although we carried out the first sensitivity study of the drug Tedizolid in Turkey, we determined the sensitivity of 10 isolates 0.063 mg/mL and 19 isolates <0,008 mg/mL. According this findings, it was determined that the sensitivity of second-line anti-TB drugs, which could'nt be studied with the MGIT method yet, was fast and reliable.
