Abstract (EN)
OBJECTIVE: The objective of this study is to search the oxidative stress (OS) which occurs in polycystic ovarian syndome whether increase or not with the treatment of clomiphene citrate (CC) that using to induce ovulation(OI) and the effect of admisitration of anti-oxidant vitamin E on possible increase. MATERIAL AND METHODS: There will be used totaly 28 rats in 4 groups of which containing 7 nuliparous young, adult, average 200-250 gr of weighted Wistar albino rats in this study. The groups are planned as: experimentally polycystic ovarian syndrommed (group I), Induced ovulation by CC with experimentally polycystic ovarian syndrommed (group II), Induced ovulation by CC with experimentally polycystic ovarian syndrommed and administered vitamin E (group III)and the control group which none of medication will be administered. (group IV)Rats wil be looked after with both 12 hours light and dark place where avaible water and instant dry food. To occur experimental polycystic ovarian syndrome in groups I, II and III; 4mg oestradiol valerat (OV) in oil solution via intramuscular injection will be applied once and after 30 days of injection it will be accepted that experimental polycystic ovarian syndrome has been occured. In Group II and III, each of the rats will be given clomiphene citrate 10 mg/kg with one injection. In Group III, additional vitamin E as intraperitoneal injection, 100mg/kg dosaged with 21days of duration. will be given with CC. Rats will be terminated by cervical dislocation on the day of 52nd under the anesthesia (Ketamine HCl 80 mg/kg and Xylazine 5 mg/kg ) and uterin horns will be taken for histological analysis. RESULTS: Under immunohistochemical analysis in group I and group II, ER? and ERß receptor concentrations are lower, SOD and iNOS levels are higher and eNOS levels are lower than group IV. In group III, with the suppliment of vitamin E, ER? and ERß receptor concentrations were increased, SOD and iNOS levels were decreased and there was an increase of the level of eNOS which was not significant. CONCLUSION: On both experimental PCOS model and CC cure which cause OS affect the receptors that play part in implantation and endometrial blood flow negatively and this process can be turn back with antioxidant vitamin E. Especially the low pregnancy rates on PCOS patients whose ovulation induced with CC may be restored with antioxidant treatment.
