Abstract (EN)
This study was planned to determine the effect of quinoa on apoptotic, autophagic, antioxidant and inflammation markers in glucocorticoid treated rats. For this reason, 42 male Wistar-Albino rats weighing 200-250 g were used. Subjects were randomly divided into six experimental groups, each consisting of 7 rats; control group (C), given to quinoa (Q), insulin resistance created (I), insulin resistance created and quinoa given (IQ), insulin resistance created and metformin given (IM), quinoa given and insulin resistance created (QI) group. Insulin resistance was established by injecting dexamethasone (1mg/kg/day) intraperitoneally for 7 days in rats in groups I, IQ, IM, QI. Group I was sacrificed after 7 days. Afterwards, quinoa feed was given to IQ for 6 days and 40 mg / kg / day metformin was given to IM for 6 days. Firstly, quinoa feed was given to QI group for 6 days and then insulin resistance (7 days) was established. The Control group was given physiological saline for 7 days and the quinoa group was given quinoa feed for 6 days. At the end of the experiment period ip. 50 g/kg/single dose of ketamine (Ketalar, Eczacibasi, Turkey) were administered to all the animals and blood and tissue samples were taken from the animals. Blood glucose, ALT, AST, ALP, total cholesterol, total protein, urea, creatinine and insulin were determined in serum samples. Liver tissues were stored at -80 ° C until the working day. For this purpose; RNA isolation products obtained from rat liver in all groups, oxidative stress (Gpx1, sod1), inflammation markers (IL1-β, TNF-α), apoptotic (Bax, Bcl2, caspase-3), autophagic (Atg5, Sqstm1), important genes involved in pathways expression was determined by real time-PCR. Glyceraldehyde-3-Phosphate dehydrogenase (GAPDH) was used as the internal control gene in real time-PCR. As a result, it has been determined that there is a decrease in the experimental insulin resistance created by dexamethasone and liver and kidney damage indicators formed by increased glucose with quinoa application. It has been determined that quinoa has significant beneficial effects on both biochemical parameters and apoptotic, autophagic, antioxidant, inflammation markers.
