The investigation of levels of anti-mullerian hormone, kisspeptin 1 and kisspeptin 1 receptor in rats

Abstract (EN)

In this thesis study, it was aimed to investigate the levels of kisspeptin 1 receptor in ovaries, Anti-mullerian hormone (AMH) and kisspeptin 1 (KiSS-1) in blood sera with cystic ovarian degeneration (COD), ovarian hyperstimulation (HS) experimentally, estrus and diestrus stages of sexual cycle in rats. For this purpose, 28 Sprague Dawley female rats aged 3-4 months were used. Rats divided into 4 groups; group 1 consisted animals in the cycle of estrus (n=7); group 2 from animals in the cycle of diestrus (n=7); group 3, animals with HS (n=7); group 4 consisted animals with COD (n=7). Animals in group 1 and 2 were decapitated when they were in estrus and diestrus cycles, group 3 and 4 were decapitated after HS and COD were created. Ovaries of animals from all groups were collected. AMH and KiSS-1 concentrations in blood sera obtained and kisspeptin 1 receptor levels in ovaries were determined using commercial ELISA kits. At the end of statistical analysis of the data obtained after the measurements revealed that the levels of kisspeptin 1 receptors on the ovary tissue were higher in estrus (1,271.43 ± 51.98 pg/ml) and HS (1,191.43 ± 85.67 pg/ml) group (p <0.05). AMH levels in blood sera were found to be higher in the group of estrus (16.91 ± 2.12 ng/ml) (p <0.05). There was no difference between the groups in terms of KiSS-1 concentrations in the blood serum. As a result, it was observed that AMH levels were lower in the experimental HS and COD cases than in the estrus phase and that the kisspeptin 1 receptor levels were similar to the estrus period in the HS rats.
