Abstract (EN)

In our study, the investigation of microorganisms with biotechnological importance in 6 different soil samples which those are, garden soil from Turkiye-Eskisehir, field soil from Turkiye-Trabzon, field soil from Yemen-Ibb, garden soil from Jordan-Amman, oil fields from Algeria-Hassi Messaoud, and gas soil from Algeria-Ain Salah. Prolonging the incubation time for bacteria from the samples was investigated and the bacteria that grew in the extended period were also isolated. The bacteria that received individual attention were isolated and subjected to morphological and enzymatic tests. On the other hand, certain microorganism groups (which fix nitrogen, use citrate as an energy source, actinomycetes and Gram-positive spore-forming) bacteria were both determined and isolated. Bacteria DNA Extraction was performed using gel electrophoresis method of bacteria that were positive in enzymatic tests. The enzymatic tests (Production of Amylase, Lipase, Proteinase, and DNase) enzymes was determined. In our study, DNA extraction, 16s rRNA, and Denaturing Gradient Gel Electrophoresis DGGE analysis were performed to identification of bacteria within the scope of culture-independent methods by amplified 16s rRNA restriction analysis (ARDRA) method. In addition to, Screening of Community Level Physiological Profiles by using Biolog Eco-Plates and also VITEK 2 as fully automated system that performs bacterial identification testing had been used.
