Investigation of the effects of sesamol on rats with cisplatin induced nephrotoxicity

Abstract (EN)
Nephrotoxicity of cisplatin (CDDP) causes oxidative stress in kidney and this restricts its clinical usage. Sesamol (5-hydroxy-1, 3- benzodioxole or 3, 4- methylenedioxyphenol) is one of the major component of sesame oil and soluble in water. In this study we investigated the effects of sesamol on CDDP induced nephrotoxicity and lipid peroxidation, experimentally. We used 28 male Wistar rats and formed the 4 groups each including 7 rarts (n=7); (i) the control group, (ii) the sesamol (8 mg/kg/gün) group, (iii) the cisplatin (7 mg/kg i.p, single dose) and (iv) the cisplatin (7 mg/kg i.p, single dose) + sesamol (8 mg/kg/gün) group respectively. Urea (p<0.05) and creatinine (p<0.05) levels were significantly higher in the cisplatin group vs the control group. In cisplatin + sesamol group; urea and creatinine levels were significantly lower than those of the cisplatin group (p<0.05). Tıssue malondialdehyde (MDA) level increased in the cisplatin group (p<0.05). Sesamol decreased tıssue MDA level significantly (p<0.05). NF-?B level increased in the cisplatin group vs the control group (p<0.001). NF-?B level decreased significantly in the cisplatin + sesamol group vs the cisplatin group (p<0.001). Nrf2 level decreased in the cisplatin group vs the control group (p<0.001) and on the other hand Nrf2 level increased in the cisplatin + sesamol group vs the cisplatin group (p<0.01). HO-1 level decrased in the cisplatin group vs the control group (p<0.001) and increased in the cisplatin + sesamol group (p<0.001). Histopathologic changes that cisplatin induced were reversed after sesamol administration. According to the result of this study cisplatin causes oxidative stress and renal injury. In conclusion in this study that sesamol reduces lipid peroxidation, induces antiinflammatory effects by reducing NF-?B level which has role on the expression of proinflammatory cytokines and has antioxidant properties by increasing the Nrf2 and HO-1 levels was determined.
