The effect of silicate applications on shoot and root development of SO4 vine rootstock under salt stress conditions in vitro

Abstract (EN)

Salinity stress is one of the most important environmental problems that limits agricultural production worldwide and affects the anatomical, physiological and enzymatic properties of plants. This study was carried out to determine the effect of potassium silicate applications on plant growth of SO4 vine rootstock under different salt stress conditions. In order to obtain the initial shoots, single-bud micro cuttings of the rootstock were cultured in MS medium containing 1 mg L-1 BAP for 3-4 weeks. The shoots formed at the end of this culture period were cultured in MS medium containing 1 mg L-1 IBA for four weeks, and the shoot tips of the rooted plants obtained were removed by cutting about 2 cm in length. Then, these shoot tips were cultured in MS medium supplemented with 1 mg L-1 IBA containing 0, 25 and 50 mg L-1 potassium silicate in combination with 0, 0.20, 0.40, 0.60 and 0.80 g L-1 NaCl. According to the results; at all salt concentrations, plant height, average root length, relative water content, membrane permeability at a dose of 50 mg L-1 of potassium silicate added to the nutrient medium; the number of nodes, plant fresh and dry weight, root fresh and dry weight, root number, SPAD index, plant damage degree, shoot and root tolerance ratio, shoot and root tolerance index and nutrient content of plant at 25 mg L-1 and 50 mg L-1 doses of potassium silicate, showed the same effect as control. Thus; it was determined that it would be appropriate to add 25 mg L-1 and 50 mg L-1 doses of potassium silicate to the nutrient medium in order to protect the SO4 vine rootstock used as a model plant in the study, from undesirable situations caused by salinity. In general, the results of this study showed that the in vitro technique can be used successfully in studies to increase the resistance and tolerance to salinity stress in existing or new breeding materials.
