Abstract (EN)

This study aimed to investigate the propagation and rooting of lily bulb scales under in vitro conditions, as well as to determine the effects of organic fertilization and different solid cultivation media on bulb development in Lilium spp. plants. In this context, 24 different nutrient media were utilized to propagate the bulb scales of Oriental hybrid Lilium spp. under in vitro conditions. The MS medium was selected as the basal medium, supplemented with varying concentrations of NAA (0.5-1 mg.l-1), BAP (0.1–0.5 mg.l-1), and TDZ (0.2–0.5 mg.l-1). Additionally, different planting methods (bulblet, scale, and leafy scale) were compared in the study. The best results were obtained in the fourth subculture. For bulblet planting, the medium containing 1.0 mg.l-1 NAA + 1.0 mg.l-1 TDZ provided the highest values in terms of leaf (37 number), bulb diameter (19.09 mm), and scale (37 number). In scales planted alone, the medium containing 0.5 mg.l-1 NAA + 0.2 mg.l-1 TDZ yielded the best results for plant lenght (40.83 mm), leaf number (11.67), bulb diameter (12.75 mm), and scale (14 number/bulb). For scales planted with a leaf segment, the medium containing 0.5 mg.l-1 NAA + 0.5 mg.l-1 TDZ produced the highest values for leaf (12.33 number) and scale (13.33 number), while the medium containing 2.0 mg.l-1 NAA + 0.2 mg.l-1 TDZ achieved the best results for bulb diameter (12.66 mm). Rooting results indicated that the best outcomes in terms of plant lenght (48.62 mm), rooting rate (100%), and root length (40.32 mm) were obtained in medium 10 (½ MS + 1 mg.l-1 IBA). Consequently, the medium containing 1 mg.l-1 NAA + 1 mg.l-1 TDZ was determined to be effective for bulblet planting, while the medium containing ½ MS + 1 mg.l-1 IBA was found to be effective for rooting.
