Abstract (EN)

In this thesis, it is aimed to evaluate the effect of nano-encapsulated drugs developed by using amphiphilic cyclodextrin nanoparticles on the lipid profile in the treatment of non-alcoholic liver lubrication (NAFL) by using Q-TOF LC / MS system in vitro. For this purpose; Various drugs currently used in treatment and nano-encapsulated formulations developed using vitamin E will be administered to the liver cancer cell line HEPG2. At the same time; drugs and vitamin E will be applied to HEPG2 cells without being encapsulated and the change in lipid profile between the two groups will be demonstrated by the C18 column using the Q-TOF LC / MS device. In the study to be carried out using the C18 column, firstly the conditions related to the analytical method will have to be optimized. For this purpose, various sample preparation techniques for the extraction of lipids from the cell line will be tried. Then, in order to separate and analyze the extracted lipids, the constant phase and mobile phase and elution program will be modified so that the highest number of lipids can be profiled. Profilable lipids will be compared semi-quantitatively for both groups and the extent to which the change is defined for each identifiable lipid. Then, by changing the lipid profile, the advantages and disadvantages of nano-encapsulated drug use will be evaluated.
