Abstract (EN)

Neurodegeneration; It is a common feature of many neurodegenerative diseases, the prevalence of which is rapidly increasing and the treatment of which is difficult. Deimination (citrullination), one of the posttranslational modifications, may occur due to inflammation, and Peptidyl arginine deiminase (PAD) enzymes are responsible for this modification. In this study, LPS was applied to the BV2 microglial cell line to create neuroinflammation and a significant increase in IL-1β, one of the cytokines, was observed. RT-PCR was performed to determine the change in PAD2, the proteins we targeted, CRP and Kininogen. A statistically significant decrease in PAD2 expression level was observed. However, there was no significant change in kininogen and CRP expression. Western blotting was performed with F95 antibody to identify deiminated proteins. A band was detected in the region where we expected CRP, and it was seen that CRP could be one of the deimination candidates. Despite the decrease in PAD expression level, deimination was identified in several protein groups. This situation is mostly seen in connection with the activity of the enzyme.
