2. ABSTRACT THE EFFECT OF REGULAR EXERCISE ON FAT METABOLISM IN ZINC PICOLINATE-SUPPLEMENTED RATS Obesity has become an increasing health problem in our country as in the world day by day. One of the most effective ways to treat obesity is regular exercise. Some minerals and supplements are also used along with regular exercise. One of these substances is Zinc picolinate. Based on this point, the aim of this study was to determine the expression levels of FAS, ZAG, ATGL, GLUT-4 and IRS1 in fatty tissues of zinc picolinate supplement in exercise-administered rats. The research was carried out after the approval of Fırat University Animal Experiments Ethics Committee (FÜHADEK) (Date: 30.05.2018, Meeting: 2018/09, Decision No: 109 and Protocol No: 2018/35). Rats used in the study were supplied by Fırat University Experimental Research Center (FÜDAM). In the study, 6 groups were formed and a total of 8-week-old male 42 Wistar albino rats were used, including 7 rats in each group. Research groups; Group I: Control, II. Group: Zinc picolinate, III. Group: Exercise, IV. Group: Exercise + Zinc picolinate, V. Group: Acute Exercise, VI. Group: Acute Exercise + Zinc picolinate. Rats were subjected to 30 minutes of jogging test for 5 days a week within 6 weeks after administration of zinc picolinate (6 mg Zn/ kg) by diet, and last day exhaustion exercise in acute exercise. As a result of applications, rats were decapitated by cervical dislocation under anesthesia and adipose tissue samples were taken. The tissues from the rats were kept at -80 °C in the deep freeze (Hettich, Germany) until they could be analyzed. The preserved samples were taken into tubes for supernatant FAS, ZAG, ATGL, GLUT-4 and IRS1 analysis and protein concentrations were determined by Western Blot (BioRad) method. The data was analyzed using the Anova procedure in the IBM SPSS (version 22) package program. Tukey Post Hoc test was used to determine the differences between the groups. Statistical significance was accepted as P <0.05. When the findings obtained in the research are evaluated; While increases in ATGL levels were observed due to chronic exercise and zinc picolinate supplementation, acute exercise did not affect ATGL levels. In terms of GLUT-4; It was determined that zinc picolinate supplement, acute and chronic exercise applications increased at GLUT-4 levels compared to the control group, and the greatest effect was in the acute exercise group with zinc picolinate supplement. In terms of ZAG parameter; It was determined that all the groups applied exercise and supplement differed from the control group, and the most effect was in the groups supplemented with exercise. When the IRS-1 variable was examined, it was determined that all groups who received exercise and supplement had an increase in IRS-1 levels compared to the control group, and the most effect was in the chronic exercise + zinc picolinate group. In terms of FAS parameter, it was observed that the control group had higher FAS levels than all other groups as a result of the practices performed. As a result of the practices, it was determined that the most effect was in the chronic exercise and chronic exercise + zinc picolinate groups. As a result, significant differences were found in IRS-1 and GLUT-4, which are markers of glucose metabolism of exercise and zinc picolinate supplement, which are administered regularly in general. Besides, regular exercise and zinc picolinate supplementation have significant effects on FAS, ATGL and ZAG values, which represent fat metabolism. For this reason, it is thought that using zinc forms together with regular exercise programs may be beneficial in order to improve the parameters related to fat and glucose metabolism.
