Abstract (EN)

Totally 100 samples were taken from patients at two Ramadi Teaching Hospitals in Ramadi. These samples were collected from patients depending on the origins of their illnesses. The isolates were obtained from wounds, ear infections, and urine infections. The 75 samples of P. aeruginosa were characterized and identified by using enrichment, selective media, and biochemical testing. The genomic DNA of 12 P. aeruginosa isolates were extracted from these various sources and the concentration and purity of these genomic DNA samples were determined to be adequate for RAPD-PCR analysis. The amplification process with the two primers produced 27 bands and 24 of which were polymorphic and ranged in size from 250 to 1500 bp. The phylogenetic connection dendrogram among 12 P. aeruginosa isolates were constructed using 24 polymorphic RAPD markers, the genetic distance, and cluster analysis among different P. aeruginosa isolates were estimated by using a Jaccard matrix of similarity and analyzed by the UPGMA computer program to supply a phylogenetic tree. The results show the RAPD-PCR analysis is a good epidemiological screening method to detect genetic variation among P. aeruginosa isolates.
