Examination of in vitro decidualization in human endometrial stromal cells

Abstract (EN)

In this study, the endometrial stromal cells in vitro 3'-5'-cyclic adenosine monophosphate (cAMP), progesterone and estrogen depending on the differentiation of decidual cells and to investigate ß-catenin and Akt signaling pathways are thought to be involved in this process with the changes in cell proliferation are intended.Experiment, in cell lines was cultured with 17-ß-estradiol, progesterone, cAMP, 17-ß-estradiol + progesterone and 17-ß-estradiol + progesterone + cAMP for three days was materialized. After morphological evaluation, differentiation was confirmed by measuring levels of prolactin hormone was expected to be synthesized by differentiated decidual cells in cell culture biochemically. As a result of morphological and biochemical investigations desidualization-specific changes occurred in the groups treated with cAMP. However, at the end of a three-day differentiation process, expression of ß-catenin was determined to be not taken place. Akt expression that is involved activation of many target proteins was viewed to be remained stable in the control and experimental groups. To assess cell proliferation, S-phase specific marker bromodeoxyuridin (BrdU) immunocytochemistry was performed. Some groups treated with cAMP that was viewed desidualization-specific changes weren?t observed any BrdU-positive cells. The rates of BrdU-positive S phase cells in the groups treated with estrogen incubation were found to be higher than the group received only progesterone.ß catenin and Akt signaling pathway not playing an active role in the early stage of desidualization process that is occurred to take part in a combination of multiple factors, S-phase specific marker BrdU-positive cell ratio that is increased by estrogen incubation not being observed in the experimental group which was created decidualization was found compatible with the fact that the suppressed proliferation in the process of differentiation.
