Abstract (EN)
Excessive weight gain and related diseases due to high-calorie intake are one of the most important health problems of today. High-calorie intake is usually due to a high carbohydrate diet. As with ketogenic diets, high-fat diets remain popular today. It is known that consumed diets which are rich in carbohydrates and fats trigger obesity and insulin resistance. In recent years, it is known that carbohydrates taken in appropriate amounts are necessary for longevity, but excessive carbohydrate restriction or excess carbohydrate intake (especially sugar) paves the way for metabolic diseases, and this is done by changing gene expressions related to metabolism. For this reason, it is important to know the change in transcription factors responsible for the modification of metabolism-related genes according to the diet. In our study, we planned to reveal the change of MONDO A and CHREBP, transcription factors which are known to directly modulate the metabolism, according to the exposed diet, Sprague Dawley rats were fed with standard, high-fat and high-carbohydrate diets during maternal and maturation periods. Afterwards, liver morphology was assessed by H&E staining, the fat accumulation of the livers was determined biochemically, and the expression levels of the transcription factors MONDO A and CHREBP were determined by immunofluorescence staining. According to the results obtained from our study, both the high-carbohydrate diet and high-fat diet caused more lipid accumulation in the liver and increased expression levels of MONDO A and CHREPB transcription factors compared to the standard diet.
