Abstract (EN)

Koşal V. Detection of sperm dna damage in benign prostatic hyperplasia model in rats. Van Yüzüncü Yil University Institute of Health Sciences, Department of Reproduction and Artificial Insemination, Ph.D. Thesis, Van, 2018. This study aimed to determine the sperm DNA damage in the benign prostatic hyperplasia model using testosterone hormone. For this purpose, male albino wistar rats, which reached an average of 200 g live weight, were used. The animals were divided into three groups of 6 animals in each group. Group 1; placebo, Group 2; 3 mg / kg daily, Group 3; Testosterone was administered intramuscularly for 30 days in a dose of 5 mg / kg daily. Sperm samples were taken from the rats at the end of the applications. Sperm DNA damage, motility, abnormal sperm rate and density analyzes were performed. By using COMET method for sperm DNA damage detection; the preparations were rated as 0 (undamaged), 1 (less damaged), 2 (damaged), 3 (moderately damaged), 4 (very damaged) and approximately 2000 spermatozoa were counted for each group. Damage rates were determined using three different methods of damage assessment. Sperm DNA damage rates according to Percentile Calculation; 1st group % 3.87454, 2nd group % 9.35852 3rd group % 15.2616, according to Arbitrary Unit; 1st group % 1.199262, 2nd group % 4.08116, 3rd group% 6.29845, according to Genetic Damage Index; group 1 % 5.5351%, group 2 % 17.0475, group 3 % 27.374 of the damage dimension was observed. This data was statistically significant and it was determined that sperm DNA damage was increased due to the amount of testosterone used (p <0.001). Motilite examination results were as follows; Group 1 %90, Group 2 %80, and Group 3 %80. A decrease in motility was observed regardless of the testosterone level (p <0.006). It was determined that all groups were not affected as a result of sperm density examination, no statistical difference was found. Abnormal sperm count results in abnormal sperm rates in group 1 (%5.75), in group 2 (%11.88) and in group 3 (%17) respectively. An increase in abnormal sperm was detected depending on the testosterone level (p <0.001). As a result; it was determined that the rate of sperm DNA damage was increased, motility and abnormal sperm ratio were adversely affected and the amount of density was not affected in the benign prostatic hyperplasia model created using the exogenous testosterone hormone. Key Words: COMET, DNA damage, sperm.
