The effect of some benzothiazole derivatives on the learning and memory parametres of streptozotocin model of alzheimer's disease in rats

In this thesis, effects of five newly synthesized benzothiazole derivatives on learning and memory parameters were investigated using streptozotocin (STZ) model of Alzheimer's disease (SMAD) in rats.Synthesis of test compounds were based on functional groups of donepezil, FK960 and sabeluzol, therapeutical drugs for AD.Plus maze, Morris water maze and avoidance tests were performed in order to examine effects of 1 mg,kg-1, 5 mg,kg-1 ve 10 mg,kg-1 doses of the test compounds on learning and memory parameters of SMAD in rats. Any effect of test compounds on spontaneous locomotor activity or anxiety level of rats were examined with the application of activity cage tests.As a result of this study, it has been revealed that, four of the five benzothiazole derivative tested compounds repaired the parameters related to the learning and memory deficits of SMAD in rats and their effects were close to the activity of reference drug, donepezile.When structure-activity relationship was taking into consideration, it was assigned that the compounds having benzyl group are more active than phenyl group in their chemical structure. Beside, more lyphophilic compounds were more active on learning and memory parameters. This result seems to be related with the easier passage of lyphophilic compounds to the central nervous system.The finding of this study are believed to light the way for the synthesis of new compounds active on learning and memory deficits of AD and contribute the drug development studies for this irremediable illness.
