Effect of exercise and metformin administration on MCT1 and MCT4, MRNA levels in muscle tissue in rats with diabetes

Abstract (EN)

The aim of this study; The aim of this study was to examine the effect of exercise applied in rats with diabetes mellitus with Streptozocin (Stz) on MCT1-MCT4 and MRNA gene expression in muscle tissue. This study was carried out on a total of 48 Wistar albino rats, 8 weeks old. Rats were randomly assigned to groups. A total of 12 rats were included in each group. All groups in the study were fed with standard water and feed. Groups; Control Group (n=12), Exercise + Diabetes Group (n=12), Metformin + Diabetes Group (n=12), Sedentary Diabetes Group (n=12). No exercise was applied to the control group. In the Exercise + Diabetes group, diabetes was created with Stz and an exercise program was applied for six weeks and five days. In the Metformin+Diabetes group, diabetes was formed with Stz, and metformin was administered by oral gavage 5 days a week. With Sedentary+Diabetes Stz, diabetes was established and no exercise was applied. At the end of six weeks, the rats were decapitated under suitable conditions and MCT1-MCT4, MRNA gene expression levels in muscle tissue were examined. In the study, descriptive statistics on blood glucose levels of the groups were examined. Control group 124.25±11.62mg/dl; Diabetes+sedentary group was 475.70±115.65 mg/dl; Diabetes+exercise group was found to be 384.48±117.36 mg/dl and Diabetes+metformin group was found to be 356.42±126.67mg/dl. When the measurement results were examined, a significant difference was determined between the blood glucose measurements between the groups (p<0.05). The Games-Howell test was used to look at the differences between the groups. It was determined that the control group had a significant difference between diabetes sedentary, diabetes exercise and diabetes metformin groups (p<0.05). There was no significant difference between diabetes sedentary, diabetes exercise and diabetes metformin groups (p>0.05). MCT1, MCT4 and MRNA gene expression levels in muscle tissue were determined by qRT-PCR and similar results were obtained in muscle tissue. Compared to the control group, the expression level of this gene decreased significantly in the metformin and sedentary groups (p<0.05), but when the exercise group was examined, it was determined that the MCT1-MCT4 and MRNA gene expression levels were higher than the control group. When MCT1, MCT4 and MRNA gene expression were examined, MCT1, MCT4 and MRNA gene expression levels were increased in muscle tissue in correlation with diabetic damage, metformin application in muscle tissue had positive effects on MCT1, MCT4 and MRNA gene expression levels, but metfromin application had MCT1, MCT4 and MRNA levels. No positive effects were found on As a result, it was understood that exercise did not affect the blood glucose level between the sedentary group and the exercise group. It was determined that exercise applied to rats had positive effects on MCT1, MCT4, MRNA gene expression levels, and Metformin application had positive effects on MCT1, MCT4, MRNA gene expression.
