Abstract (EN)
In this study, it was aimed to determine the general composition of protein and protein concentrate obtained from carob embryo waste and to determine the bioactive properties (ACE inhibition and antioxidant activity) of protein hydrolysates obtained from carob embryo waste. According to the results obtained, carob embryo contains 24.26% protein, 7.29% moisture, 5.08% ash, 4.17% fat and 59.2% carbohydrates. Carob embryo protein concentrate was found to contain 80.72% protein, 0.4% moisture, 4.06% ash, 0.38% fat and 14.34% carbohydrates. Antioxidant activity of protein hydrolysates obtained from carob embryo waste was found using Frap and DPPH methods. According to the results of the DPPH method obtained, P0 and T0 samples showed antioxidant activity of 1.03 mgtrolox/g (dry matter) when hydrolyzed with trypsin enzyme (T30, T60, T120), respectively. ,48 mgtrolox/g (dry matter), 1.53 mgtrolox/g (dry matter), 1.58 mgtrolox/g (dry matter), when hydrolyzed with pepsin enzyme (P30, P60, P120), respectively 1.10 mgtrolox/g(dry matter) 1.11 mgtrolox/g(dry matter), 1.18 mgtrolox/g(dry matter). Antioxidant activity According to the results of the Frap method, P0 and T0 samples showed antioxidant activity of 6.63 mgtrolox/g (dry matter) when hydrolyzed with pepsin enzyme (P30, P60, P120), respectively. 7.89 mgtrolox/g (dry matter), 8.07 mgtrolox/g (dry matter) and 9.92 mgtrolox/g (dry matter) when we hydrolyze it with trypsin enzyme (T30, T60, T120, respectively), It appears to be 9.81 mgtrolox/g (dry matter), 9.67 mgtrolox/g (dry matter) and 10.59 mgtrolox/g (dry matter). According to the results obtained in ACE inhibition of protein hydrolysates obtained from carob embryo waste, the ACE inhibition (%) values of P0 and T0 samples taken before hydrolysis was 87,78 P30, P60, P120 ACE inhibition (%) obtained as a result of hydrolysis with pepsin enzyme values vary between 90.26-91.52. T30, T60, T120 ACE inhibition values obtained as a result of hydrolysis with trypsin enzyme (%) ranged from 88.67 to 90.38. IC50 values were checked to find out which hydrolysates and protein were more active. The P0 value for pepsin hydrolysates based on IC50 values is 2.63 mg protein/ml. P30, P60 and P120 values are 0.76 mg protein/ml, 0.76 mg protein/ml and 0.70 mg protein/ml, respectively. The T0 value for trypsin hydrolysates is 2.63 mg protein/ml. T30, T60 and T120 values were found to be 0.91 mg protein/ml, 0.88 mg protein/ml and 0.78 mg protein/ml, respectively.
