Abstract (EN)
Obesity is one of the main factors spreading rapidly all over the world and underlying many diseases. Today, the endocrine functions of peripheral tissues gain importance in the guideline of strength homeostasis. Irisin, adropin then preptin are newly discovered endocrine factors involved in the guideline of strength homeostasis. Irisin is a muscle-derived protein and reasons energy spending by altering white adipose tissue to brown adipose tissue. Adropin is a protein encoded via the ENHO gene, which acting a role in regulating strength homeostasis and keeping insulin response. Preptin is a peptide hormone absorbed together through insulin from the β cells of the pancreas. It is thought that these energy regulatory peptides may provide clinical benefit by playing a role in cellular communication in the fight against and T2DM. Our study consists of individuals aged 18-50 years, who are overweight or obese (n= 59) , healthy (control, n=59) and T2DM (n= 59) . All blood samples were taken in to gel tube, centrifuged for 10 minutes at 3500 rpm by the side of 4°C. The serum portion stayed separated into Eppendorf tubes containing aprotinin, a protease inhibitor, to be studied, and stored in a deep freezer at -20°C until the study day. After the blood collection was completed, Irisin, adropin and preptin levels will be analysed in blood serum samples, using commercially obtainable ELISA kits. The amount of irisin, adropin and preptin was examined in serum samples prepared using a commercially available ELISA kit. İRİSİN (NG/ML) in healthy, obesity and obesity patients with type 2 diabetes mellitus were result show that level of İRİSİN in the obesity groups and T2DM groups are significantly decrease than the control group (p <0.05, F =19.617). ADROPİN (PG/ML) in healthy, obesity and obesity patients with type 2 diabetes mellitus were result show that level of ADROPİN in the obesity groups and T2DM groups are significantly decrease than the control group. (p <0.05, F =3.632). PREPTİN (NG/L) in the three groups of control, obesity and T2DM were result show that level of PREPTİN significantly increase in obesity and patients with Type2 diabetes mellitus compered to the control group (p <0.05, F =6.846). With this work; It was aimed to investigate the relationship of these three endocrine factors with energy homeostasis by comparing the concentrations of irisin, adropin and preptin in obese ,T2DM and non-obese groups. The fact that all three of the parameters planned to be looked at are related to energy metabolism and that obesity is an increasingly important health problem has been effective in the planning of this study. After comparing these parameter we found that serum irisin , adropin and preptin should be utilized as a biomarker for assessing the risk of developing T2DM2and control obesity.
