Investigation of the effectiveness of thrombocyte-RİCH plasma in the treatment of osteochondral lesions: Experimental rat model

Abstract (EN)
Objective: In the present study, it was aimed to clinically and radiographically monitor the effectiveness of platelet-rich plasma (PRP) injection in the treatment of osteoarthritis in the experimental model created by intra-articular injection of monosodium iodoacetate. Material and Methods: To create OA in the experimental model; In a total of 24 rats, 3mg sodium iodoacetate solution in 50 microliter saline was injected into the right genu joint under general anesthesia with intraperitoneal 8 mg/kg xylazine + 60 mg/kg ketamine under aseptic conditions. The genu joint of the rats was followed clinically and radiologically for two weeks. As a result of the development of OA, the rats were randomly divided into PRP (12 rats) and control (12 rats) groups. The rats in the PRP group were injected intraarticularly with 0.15 ml PRP into the right genu joint. The control group was injected with 0.15 ml of saline. Clinically, changes in hind leg posture were observed on the 15th, 30th, 45th and 60th days before and after monosodium iodoacetate administration in both groups, knee circumference and body weight gain were recorded. X-rays of the right genu joints of the animals in all groups in Anterior/Posterior (AP) and Medio/Lateral (ML) positions were taken and scored on the 15th, 30th, 45th and 60th days before and after the experiment. Results: In the statistical analysis, Leg Posture Score (BPS) showed that there was no difference between the groups in periodic measurements. It was observed that the knee circumference measurements increased in favor of the PRP group. While the live weight gain was significant between the groups in the first 2 measurements, there was no difference in the following periods. Narrowing in the joint space and the development of sclerosis in the subchondral bone showed significant time in favor of PRP (p<0.0001). Osteophyte development from achoson did not differ between groups. Conclusion: As a result, monosodium iodoacetate was effective for the formation of OA in the experimental model; It has been concluded that PRP application may be beneficial in the treatment of OA in the light of clinical and radiological data, but it should be followed with advanced imaging methods and histochemical markers to reach a definitive conclusion.
