The effect of astaxanthin in the ovary of streptozocin-induced TYPE 1 diabetes rats

Abstract (EN)
Aim of the Study : To determine the effect of Astaxanthin on the damage caused by diabetes in the ovary, and to determine the change in this effect when Astaxanthin is used together with or without Insulin. Histological effects, autophagic immunohistochemical markers, and biochemical oxidation parameters of astaxanthin were examined. Material and Method : In our study, 5 groups with 6 animals in each group were formed: Control, Diabetes, Diabetes+Astaxanthin, Diabetes+Insulin, Diabetes+Astaxanthin+Insulin. Intraperitoneal 50mg/kg Streptozocin injection was performed in all groups with diabetes. After the first 4 weeks of the experiment were completed, 25 mg/kg Astaxanthin was administered by oral gavage to each subject in the D+A group for 7 days during the 5th week, 0.5 IU/kg Insulin i.p was administered to each subject in the D+I group. 25 mg/kg Astaxanthin was given by oral gavage and 0.5 IU/kg Insulin was given intraperitoneally to each subject. Results : Hematoxylin-Eosin (H-E), Masson Trichrome, ATG5 and LC3 antibodies were stained for histopathological and immunohistochemical evaluations in the ovaries collected from the subjects. It was observed that the incidence of cystic follicle, atretic follicle, degenerated granulosa cell, CL degeneration, fibrotic and hemorrhagic medulla increased in Diabetes compared to the control group, and was closer to normal in the treatment groups, especially in D+A+I. It was determined that TAS and GSH values were lower in the Diabetes group compared to the control group, the values in the treatment groups were closer to normal, and the MDA values were increased in the Diabetes group compared to the control group, but they were closer to normal values in the treatment groups. There were no significant difference between groups for ATG5 and LC3. Conclusion : Astaxanthin has the potential to be an effective alternative or a combination to insulin in reducing the damage done by Type I Diabetes on the ovary through oxidation. Experiments of different doses and different living species are required on the subject.
