Abstract (EN)

Male infertility is the condition of a man not being a natural father. Male infertility has two characteristics: sperm deficiency and morphological anomalies. Although leptin is necessary for healthy reproductive function; Excessive amounts of leptin can have harmful effects on the male reproductive system. In this thesis, it was aimed to determine whether the differences in weight, sperm morphology and motility of male offspring of 10-week-old Balb-C mice whose mothers were injected with subcutaneous leptin during pregnancy are associated with infertility. The effects of leptin treatment on the weight, sperm morphology and motility of males in the second generation of mice were examined. After establishing a pregnancy in 12-week-old Balb-C mice, the group that received subcutaneous leptin injection was designed as the experimental group. The group that received subcutaneous distilled water injection was designed as the control group. Epididymis of second generation 10-week-old male Balb-C mice were removed and the sperm samples obtained were compared with the control group. To determine the sperm morphology, sperm counts were performed on the sperm samples stained with Spermac at 100X magnification under a light microscope. To determine sperm motility, motile and immobile sperm counts were made using a Makler camera at 40X magnification. When the weights of the control and experimental groups were evaluated, no significant difference was observed between the groups (p>0.05). An increase in the curved tail anomaly in the experimental group compared to the control group; decrease in normal sperm was found to be statistically significant (p<0.05). A decrease in the number of immobile sperm between the experimental group and the control group (p<0.05); a statistically significant increase was determined in the motile and total sperm counts (p<0.05). Leptin is among the first datasets showing the sperm motility and morphology of the second generation. In addition to these studies with leptin, a study claiming that second-generation male infertility is related to first-generation leptin will be among the first in the scientific literature.
