Abstract (EN)
Renal ischemia-reperfusion injury (I / R), the molecular steps are not fully understood, is a pathophysiological processes. The degree of ischemic damage, ranging from molecular step in cell death although it depends on the depth of hypoxia triggers. In the study, we aimed to evaluate the effects of verapamil, a Ca++ channel blocker, and lidocaine, a Na+ channel blocker, on the genes play role in autophagy pathway in the kidney cells subjected to the I/R injury. For this, 5 autohagy genes; Atg5, Atg7, Atg10, Becn1 and Ulk1 were selected. 24 adult Wistar-albino rats of weight 200–250 grams were used. Rats, after anesthesia along the midline laparotomy was performed clamping the renal artery and vein. Gruops and features; Group I= Sham; not subjected to ischemia/reperfussion, Group II= I/R group; After ischemia reperfusion for 60 minutes was performed 48 hours. Group III = I / R, and calcium (Ca ++) channel blockers,the group:30 minutes prior to anesthesia verapamil (Ver) by oral 1.25 mg / kg doses were given pre-treatment, after I / R was performed. Group IV= lidocaine (L) (1 mg/kg) was intraperitoneally given to the rats as pretreatment in this group 30 minutes before anesthesia, then the rats were subjected to I/R. The advanced molecular analysises (RNA isolation, cDNA synthesis and qPCR) were perfomed to the obtained tissues. The expressions of Atg5, Atg7, Atg10, Becn1 and Ulk1 genes were statistically increased in the I/R, I/R Ca++ (group III) and I/R Na+ (group IV) groups compared to the sham. Also, a significant decrease occured in the all genes in I/R Ca++ group compared to the between I/R groups.
