Abstract (EN)

Ischemia is defined that deprived of oxygen outcome of insufficient perfusion, depending upon decline of blood stream. The paradoxally reperfusion of ischemic tissue cause more critical damage as induced by ischemia. The lung damage with induced by Ischemia / Reperfusion (I/R) is frequently encountered situation in lung transplantation. In our study, expression of TRPM channels in lung tissues, after I/R model formed, were monitored. Thus, totally a number of 40 Winstar Albino rat were equally seperated in 4 groups. Groups were defined as Group I- control group, Group II= 1 hour schemia, Group III= 1 hour schemia + 2 hours reperfusion, Group IV = 1 hour schemia+ 4 hours reperfusion groups. RNA isolation, cDNA synthesis and RT-PCR (Revers Transkriptaz-PCR) were carried out. Moreover, lung tissues were studied histapatologically. In the end of the study we carried out, it was observed that, expressions of all channels increased in schemia group, while no expression was observed in İ/R 1 group and was observed anew increase in I/R 2 group. In this situation, superoxides accumulated in schemia is thought to harm the tissue in reacting with various components, yet in İ/R 2 group it is thought that the tissue itself is stuggling to recover, superoxides are decreasing in the environment and the cell is trying to make itself stable.
