Investigation of the expression levels of GALNT5 and ZDHHC9 genes in right and left colon tumors

Abstract (EN)

Colorectal cancers are in the third place in terms of incidence in both genders. In colon cancer, clinical, pathological and genetic differences occur depending on whether the tumor is located in the right or left colon. This situation can change the course of the disease and the treatment method to be applied. In our study, we examined the expressions of ZDHHC9 and GALNT5 genes in tumoral tissues located in the right and left colon. Our sample; It consisted of 89 tumor tissue samples, 42 of which were right colon and 47 left colon, and healthy tissue samples of the same individuals adjacent to the tumor tissue in Mersin University Faculty of Medicine, Department of Pathology. Our work; performed on archival materials embedded in a paraffin block prepared from tissues with adenocarcinoma, the most common type of colorectal cancer. Before the RNA isolation from the tissue samples, cDNA synthesis was performed. Expression of ZDHHC9 and GALNT5 genes was analyzed in Real Time PCR by comparative CT (ΔΔCT) method. Statistical evaluation of the results was done with Wilcoxon T-test, Student's T-test and Mann-Whitney U test. Colorectal cancer is more common in males than females (p<0.05). In general, when evaluated separately as right and left colon tumors: There is an increase in the expression levels of both ZDHHC9 and GALNT5 genes (p<0.001). When we compared the expression of both genes in right and left colon tumors, GALNT5 gene expression in the left colon was found to be higher than in the right colon (p<0.048). The fact that the expression of ZDHHC9 and GALNT5 genes is higher in both right and left colon tumors indicates that they may be risk factors for developing colon cancer. The fact that the left colon GALNT5 gene expression is higher than the right colon can be an important indicator in explaining the fact that left colon cancers are more common than right colon cancers. This study, which we conducted on the basis of Mersin sample, is the first study in our country on the expression values of ZDHHC9 and GALNT5 genes in colorectal cancer tissue and control groups.
