Abstract (EN)

Endometrial cancer is the first type of cancer among gynecological cancers in women. Studies on diagnosis and treatment phase have gained importance recently. In addition to existing treatment options, traditional medicines are being developed. Resveratrol is a phytoalexin with many biological effects such as anti-inflammatory and anticancer effects. Resveratrol is thought to exert its therapeutic effect in cancer through cell death mechanisms. However, the mechanism of these effects is not fully understood. In this study, it was aimed to investigate the dose-dependent effects of Resveratrol on Ishikawa cells, an endometrial cancer cell line. The cytotoxic effect of resveratrol on Ishikawa cells was determined by MTT test. mRNA Expression Analysis of Apoptosis and Autophagy Associated Genes by RT-qPCR with IC50 dose was performed. After the IC50 dose was applied, immunofluorescent staining was performed. Resveratrol appeared to inhibit the proliferation of Ishikawa cells in a dose-dependent manner. When the IC50 dose group was compared with the control group cells according to their mRNA expression levels, a significant increase was observed in the levels of Beclin-1, LC3-I, LC3-II, Casp-7, Casp-8, Casp-9, CycS and FADD. In our study, Resveratrol's induction of apoptosis and autophagy suggested that it may be an agent that can be used in cancer treatment.
