Abstract (EN)

The aim of this study; This study includes biochemical measurements on blood glucose levels and CGRP and LDH gene expression in liver and muscle tissue after heavy exercise and metformin administration in rats with streptozocin (STZ) causing diabetes. Four sets of 12 Wistar albino male rats were used in this study. Standard water and food were given to each group during the trial. Groups: Control Group (12 members), Exercise + Diabetes Group (12 members), Metformin + Diabetes Group (12 members), and Sedentary Diabetes Group (12 members). The control group did not engage in strenuous activity. Diabetes was induced with STZ in the Exercise + Diabetes group, and for six weeks, a rigorous exercise regimen was implemented five days per week. Diabetes was induced with STZ in the Metformin + Diabetes group, and metformin was given orally five days per week. Diabetes + Sedentary Lifestyle Diabetes was diagnosed with STZ and no exercise was prescribed. The rats were decapitated under controlled circumstances after six weeks, and the CGRP and LDH gene. Diabetes was induced with STZ in the Metformin + Diabetes group, and metformin was given orally five days per week. Diabetes + Sedentary Lifestyle Diabetes was diagnosed with STZ and no exercise was prescribed. At the end of 6 weeks, the rats were decapitated under suitable conditions and the CGRP and LDH gene expression levels in the liver and muscle tissue were examined. In the study, descriptive statistics of the groups were examined. The control group was 87.75±6.30; Diabetes+sedentary group 388.26±175.99; Diabetes+exercise group 388.26±217.36 and Diabetes+metformin group 351.77±186.67. When the measurement results were examined, it was observed that there was a significant difference between the groups in terms of blood glucose measurements (p<0.05). The Games-Howell test was used to look at the differences between the groups. It was observed that there was a significant difference between the control group with diabetes sedentary, diabetes exercise and diabetes metformin groups (p<0.05). There was no significant difference between diabetes sedentary, diabetes exercise and diabetes metformin groups (p>0.05). CGRP gene expression levels in liver and muscle tissue were determined by QRT-PCR, and similar results were obtained in liver and muscle tissue. Compared to the control group, the expression level of this gene decreased significantly in the metformin and sedentary groups (p<0.05), but when the exercise group was examined, it was determined that the CGRP gene expression level was higher than the control group. When LDH gene expression was examined, it was observed that LDH gene expression levels increased in both muscle tissue and liver tissue in correlation with diabetic damage, metformin administration in muscle tissue had positive effects on LDH gene expression levels, but metformin administration in liver tissue had positive effects on LDH levels. When the study findings were examined, it was understood that heavy exercise did not affect the blood glucose level between the sedentary group and the exercise group. It was determined that the exercise applied to the rats had positive effects on the CGRP gene expression level, and the Metformin application had a positive effect on the LDH gene expression. It was concluded that strenuous exercise has an ameliorative effect on CGRP and LDH gene expression.
