The releationship between micro RNA (miRNA) expression levels and response to treatment in hematologic malignancies

Abstract (EN)
Hematologic malignancies arise as a result of uncontrolled increase of stem cells that are responsible for blood cell production. Involved in the epigenetic regulation of microRNA (miRNA) molecules that play a role in cellular events such as cell proliferation, cell differentiation, organogenesis, apoptosis, and development of cancer, heart disease and various infectious diseases. MicroRNAs have a critical role in the regulation of hematopoiesis and show oncogenic or tumor-suppressive effect in hematological cancers. In studies, there is evidence about various miRNA genes' expression levels are differences before and after treatment of patients in hematologic malignancies. However, different studies are needed with large patient populations. In this study, expression levels of miR-17-92 gene cluster it covers miR-17, 18a, 19a, 19b, 20a and miR-92a in hematologic malignancies and the association between treatment response is analyzed. miRNAs expression levels obtained from the Blood samples were taken at pre and post-treatment from 24 patients that were diagnosed with Hematologic malignancies of acute myeloid leukemia, acute and chronic lymphoblastic leukemia, Hodgkin's and Non-Hodgkin's lymphoma and 33 healthy blood samples determining by a method of the hybridization-based micro series and compared statistically. Evaluation of the results of the samples before and after treatment of patients, miR-17-92 gene cluster expression levels compared to control group members were found to have increased (p=0.001). In comparing the values before and after treatment of mir-19a and 19b, the post-treatment group was found higher (p=0.001). Analyzed values before and after treatment of patients who respond to treatment and not also miR-19 and miR-19b were increased in responding to treatment group (p=0.001, p=0.009). As a result of miR-17-92 gene cluster members miR-19a and miR-19b expression levels increased after treatment so that the molecular theraphy to be conducted for research that may lead.
