In vitro investigation of the effects of WNT inhibitor PRI-724 on pancreatic cancer stem cells

Abstract (EN)

Objective: In this study, it was aimed to evaluate the effect of Wnt inhibitör PRI-724 on pancreatic cancer stem cells. Materials and Methods: Our research was conducted in the Laboratory of Basic Medical Sciences of Adnan Menderes University. Our research started with a literature review in December 2020. April 2021- September 2021 experimental groups were created and MTS/PMS dose determination, Flow Cytometry and Clonogenic Assay were performed with the obtained cells. Results: A dose determination experiment was performed to determine the IC50 value of the PRI-724, Gemcitabine and Combined (Gem+PRI-724) groups. As a result of this experiment, the IC50 value of PRI-724 was determined to be 45 µM, the IC50 value of Gemcitabine was determined to be 4 mg/ml, and the IC50 value of the combined (Gem+PRI-724) group was determined to be 4mg/ml+45 µM. In the Flow Cytometry experiment, the control, Gemcitabine, PRI-724 and Combined (Gem+PRI-724) groups were created. By adding CD44, CD133 and CD24 antibodies to each group, scattering graphs were obtained and then scattering graphs were evaluated. In the clonogenic experiment, control, gemcitabine, PRI-724 and combined (Gem+PRI-724) groups were created. While it was observed that cells formed colonies in the control group, it was observed that they did not form colonies in other groups that were administered the drug. Conclusion: According to the results of our study, it was observed that the Wnt inhibitör PRI-724 reduces pancreatic cancer cells by inhibiting pancreatic cancer stem cells. It is expected that this study may be the hope in the future for the treatment of pancreatic cancer and may lead to clinical trials.
