Abstract (EN)

When the nutrition of the newborn is examined historically, the most important nutrient is breast milk. Breast milk is an easily digestible food with high bioavailability, containing fluid, energy and nutrients required for optimum growth and development of the newborn baby. Gastric cancer is a cancer type with a high incidence and mortality rate all over the world. While gastric cancer, dietary factors, adequate and healthy nutrition, diets rich in fiber, early diagnosis, and genetic factors decrease the incidence of gastric cancer, excessive consumption of tobacco products and alcohol increases the incidence of gastric cancer. In this study, the effect of whey proteins obtained from breast milk, which taken from 10 different mothers having 0-6 month old babies, on metastasis was investigated. It has been studied through the cell migration assay of breast milk whey proteins on metastasis and gene expression of PPARRγ, which has been associated with metastasis in various types of cancer. In our study, gastric cancer cells were grown in RPMI medium at 37oC and 5% CO2 conditions. Proliferating cells were seeded in 12-well cell culture dishes at 200,000 cells/well and the cells were confluent. Then, 50 µL of whey proteins obtained from breast milk were applied to each well, and then a straight scratch was made with a pipette tip. It were photographed at 0 and 24 hours under the microscope to detect cell migration. Total RNA was isolated from cells for PPARγ gene expression analysis and the effect of breast milk whey proteins on PPARγ was analyzed by RT-PCR. As result of the study, it was determined that 2nd, 3rd, 5th and 8th breast milk whey proteins suppressed metastasis, however 6th, 7th, 9th and 10th breast milk whey proteins didn't suppress metastasis. Conversely, no closed was observed in gastric cancer cells given 1st and 4th breast milk whey proteins, but an increase in the width of the scratch was observed. While PPARγ gene expression increased significantly (p<0.05) in the cells to which 3rd, 4th, 5th, 6th and 7th breast milk whey proteins were applied, the gene expression change was not significant in the others. As a result, since the whey protein profile of each breast milk is different, the effect of breast milk whey proteins on gastric cancer should be investigated with PPARγ and different metastatic pathways.
